I know there's been a lot of misgivings expressed regarding S6. I'm not really one way or the other about it right now; I'm just trying to get through this season.
But the fact that
Jensen is excited and looking forward to S6? Yes, that makes me excited.

Because it will be headed up by different writers and be something of a new experience, with completely new stories, within the context of the old experience. (I hope that made sense.) If nothing else, he gets to put food on the table for another year.
(I loved Jim Beaver's response to the renewal, which was to thank WB and the CW for allowing him to feed his child for another year.)

As a viewer, when I see series that have gone on as long as Supernatural I get the feeling that the actors get kind of bored and phone in the performance simply because they know the character so well. I imagine that as an actor Jensen is excited because he has no clue where Dean is going to be in season 6, which means things will be a little more challenging. As a fan, anything that keeps the cast and crew motivated to do good work is a positive in my book.

He gets tired of being "stuck" (not the word he wanted to use) in Vancouver, but otoh, he gets to work at a job he loves with people he loves to work with. I have no way to read Jensen's mind, but I think the change in next year's storytelling is going to do a lot of good for that burnt out feeling. If it had just been a continuation of this season, I think he might not be as excited as he seems to be.
I've also wondered if S6 is why he proposed to Danneel when he did. If he got tired of waiting or whatever or just wanted her up there as his wife, rather than sporadically as his girlfriend. (Of course, that's all pure speculation on my part. For all I know, she'll be maintaining a house in L.A. while he's up in BC).
JMO. YMMV.
But, yeah, as long as he's happy working and I'm happy watching, it's all good. :-)
So I think you're very right in that Jensen is probably quite anticipating the unknown of S6.
I've also gotten the distinct impression that he likes when the brothers are fighting evil together, back to back, "saving people, hunting things; the family business." And if the indicators are that they're going to get back to that formula, I think he's probably liking that idea, too.
The thing about Jensen that I've observed, is that I don't think he's ever really "insincere." Snarky, wry, sarcastic, yes. But what I've observed of him from my little corner of the internet and the fandom is that he's generally honest in his responses. Ya know?
